Quantum BioPharma Taps Thermo Fisher to Launch Phase 2 Lucid-MS Trial in Progressive MS

Quantum BioPharma Ltd. (NASDAQ: QNTM) has taken a critical step toward advancing its lead multiple sclerosis drug candidate, Lucid-MS, by signing a Study Start-Up Agreement with the clinical research business of Thermo Fisher Scientific. The agreement, announced this week, moves the program from planning into execution and sets the stage for a Phase 2 trial in progressive forms of MS, where treatment options remain severely limited. The partnership matters because it pairs a first-in-class neuroprotective approach with one of the world's most experienced contract research organizations, potentially accelerating a therapy that targets the disease's underlying cause rather than just its symptoms.

Quantum BioPharma (NASDAQ: QNTM), a biopharmaceutical company focused on neurodegenerative and metabolic disorders, signed the agreement with Thermo Fisher's clinical research business, a global CRO that the company acquired in 2021 for $17.4 billion. The contract covers implementation of the Phase 2 trial of Lucid-MS, which is designed to evaluate efficacy, safety, and tolerability in people with progressive MS. The trial will be randomized, double-blind, and placebo-controlled, assessing clinical and radiological endpoints tied to disease progression. The move follows the U.S. Food and Drug Administration's clearance of Quantum's Investigational New Drug application for Lucid-MS in August, clearing the way for human testing.

Multiple sclerosis affects an estimated 3 million people worldwide, including nearly 1 million in the United States. The disease damages myelin, the protective coating around nerve fibers, leading to a range of disabling symptoms. While most existing MS drugs work by modulating or suppressing the immune system, Lucid-MS takes a different tack: it aims to protect the myelin sheath itself. The drug is an investigational, first-in-class new chemical entity that targets peptidyl arginine deiminase 2 (PAD2), an enzyme implicated in the hypercitrullination of myelin proteins. By inhibiting PAD2, Lucid-MS may prevent or even reverse myelin degradation, a mechanism that preclinical models have supported. In Phase 1 trials in healthy participants, the drug showed a favorable safety profile and was well tolerated.

The need for new approaches is acute in progressive MS. A 2023 study in Neurology and Therapy described treatment options for progressive MS as limited, with many patients remaining untreated or continuing to decline despite available therapies. A National MS Society report released this year, drawing on survey responses from more than 2,800 people living with MS, identified urgent unmet needs, including effective treatment of disease progression and therapies that repair myelin damage. Quantum's neuroprotective strategy directly addresses these gaps, and the Phase 2 trial will provide the first rigorous test of whether the approach translates into clinical benefit.

Thermo Fisher's clinical research business brings deep expertise in central nervous system and MS trials. "We're proud to partner with Quantum BioPharma in advancing their neuro-degenerative clinical program in multiple sclerosis," said Michael Cioffi, vice president and general manager, neuroscience, clinical research, at Thermo Fisher. "Our team brings deep experience in CNS and MS trials, along with an integrated approach that connects strategy and execution to help deliver these complex studies." Quantum's CEO, Zeeshan Saeed, called the agreement an important next step following FDA clearance, while Andrzej Chruscinski, Quantum's President of Scientific and Clinical Affairs, noted that the Phase 2 study will allow evaluation of the innovative approach using clinical and radiological measures.

The commercial stakes are substantial. Quantum's announcement cites Grand View Research in projecting that the MS therapeutics market will exceed $38 billion by 2030. If Lucid-MS succeeds, it could capture a significant share of that market by offering a novel mechanism for patients who currently have few options.
